【中文摘要】 目的:探讨丹参对内毒素(LPS)致急性肺损伤(ALI)大鼠转化生长因子β_1(TGF-β_1)表达的影响。 方法:用LPS(E.Coli O_(55):B_5)复制ALI大鼠模型。健康SD大鼠24只采用简单随机的方法分为3组,每组8只,雌雄不拘。1)ALI组:LPS(5mg/kg)经尾静脉推注;2)对照组:LPS用生理盐水代替;3)丹参治疗组:LPS(5mg/kg)经尾静脉推注,1h后经尾静脉推注丹参(10g/kg)。各组动物模型制备后均于LPS或生理盐水注射后观察12h,观察有否呼吸困难及呼吸频率变化。取静脉血2ml抗凝后,用血液粘度测试仪进行血流变分析。放血处死动物,然后开胸取整个左肺,用滤纸吸干血渍,分析天平称重后放入80℃恒温烤箱烘烤24小时,计算肺湿/干重(W/D)比值来评价肺组织的水肿程度。右肺上叶组织制备病理切片,进行HE染色和免疫组化染色,光镜下观察组织病理学变化及TGF-β_1的染色结果,并用H score方法对TGF-β_1的表达进行半定量评价。 结果:1、肺组织病理切片光镜观察:ALI组肺组织毛细血管充血,肺间质肿胀,肺泡间隔水肿增厚,内有大量炎性细胞浸润... 【英文摘要】 Objective: To investigate the effect of Radix Salviae Miltiorrhizae on transforming growth factor beta1(TGF-β_1) in rats with LPS-induced acute lung injury.
