【中文摘要】 目的:观察海带多糖L01对血小板功能和血管内皮细胞的体内保护作用以及对体外培养内皮细胞分泌血管性假血友病因子(vWF)的影响,为研究和开发新型抗血栓药物提供理论和实验室依据。 方法:(1)体内给药对血小板粘附聚集功能的影响。采用大剂量肾上腺素(Adr)法制备内皮细胞损伤的大鼠模型。大鼠随机分为4组,模型组、L01高、低剂量组和生理盐水对照组,分别给药1d、2d、3d、4d和5d。模型组注射Adr并予以生理盐水治疗;L01高、低剂量组分别注射等剂量Adr,予以高、低剂量L01治疗;生理盐水组则注射等剂量生理盐水,予以等剂量生理盐水治疗。腹主动脉取血,采用滤过法观测各组大鼠血小板粘附率,火棉胶-氯仿玻片法观测血小板聚集率。(2)体内给药对内皮细胞分泌vWF的影响。各组大鼠取血后,用免疫组织化学方法特异性地显示胸主动脉内皮细胞中的vWF,使内皮染色,观察血管内皮的损伤程度,并用ELISA法检测大鼠血浆中vWF的浓度。(3)对体外培养内皮细胞分泌vWF的影响。体外培养的人脐静脉内皮细胞(HUVEC)接种在24孔板,随机分为空白对照组、模型组、L01对照组、L01高剂量组、L01低剂量组,分别培养24...
